摘要:
目的 探讨阿托伐他汀对胰岛素抵抗(IR)小鼠心室重构的影响及机制。方法 采用随机数字表法将40只C57BL/6野生型雄性小鼠分为4组:正常对照组(Control组)、IR组、Ato10组、Ato20组,每组10只。Control组给予普通饲料喂养,其余3组均给予高脂饲料喂养。Ato10组和Ato20组同时每日分别给予阿托伐他汀10mg·(kg·d)-1和20mg·(kg·d)-1灌胃,连续喂养干预12周。检测空腹血糖(FPG);ELISA法检测血清空腹胰岛素(FINS)、脂联素(APN)及转化生长因子-β1(TGF-β1)表达水平;超声心动图检测左心室后壁厚度(LVPWd)、室间隔厚度(IVSd)、左心室射血分数(LVEF)及左心室短轴缩短率(LVFS);称量小鼠体质量、全心质量及左心室质量;HE染色观察小鼠心肌组织结构的变化;免疫组织化学法和Western blot检测心肌组织中APN和TGF-β1表达水平。结果 与Control组比较,IR组LVPWd和IVSd增加,LVEF和LVFS降低,FPG、FINS及HOMA-IR升高,体质量、全心质量及左心室质量升高,心肌细胞排列紊乱,肌纤维扭曲变形且发生断裂,血清和心肌组织APN表达下降,TGF-β1表达升高(P均<0.05);与IR组比较,Ato10组LVEF和LVFS升高,FPG和HOMA-IR下降,心肌微观结构变化改善,血清和心肌组织APN表达升高,心肌组织TGF-β1表达降低(P均<0.05),Ato20组LVPWd和IVSd减小,LVEF和LVFS升高,FPG、FINS及HOMA-IR降低,体质量、全心质量及左心室质量降低,心肌微观结构变化进一步改善,血清和心肌组织APN表达升高,TGF-β1表达降低,且Ato20组改善效果更显著(P均<0.05)。结论 IR可导致小鼠心室重构,阿托伐他汀可以改善IR所致的心室重构,且存在剂量依赖性,推测其机制可能与APN表达水平升高有关。
Abstract:
Objective To investigate the effect and mechanism of atorvastatin on ventricular remodeling in insulin resistance (IR) mice.Methods Total 40 C57BL/6 wild-type male mice were divided into 4 groups by random number table: normal control group (Control group), IR group, Ato10 group and Ato20 group with 10 rats in each group.The Control group was fed with normal diet, and the other three groups were fed with high-fat diet.The Ato10 group[10mg·(kg·d)-1)] and the Ato20 group [20mg·(kg·d)-1)] were given atorvastatin every day for 12 weeks.Fasting blood glucose (FPG) was measured.Serum fasting insulin (FINS), adiponectin (APN )and transforming growth factor-β1 (TGF-β1) were detected by ELISA.Left ventricular posterior wall thickness(LVPWd), interventricular septal thickness(IVSd),left ventricular ejection fraction (LVEF), and left ventricular short axis shortening rate(LVFS) were measured by echocardiography.Mouse body weight, whole heart weight and left ventricular weight were measured.HE staining was used to observe the changes of myocardial tissue structure in mice.The expression levels of APN and TGF-β1 in myocardium were detected by immunohistochemistry and Western blot.Results Compared with the Control group, the LVPWd and IVSd were increased, and the LVEF and LVFS were decreased.FPG,FINS and HOMA-IR were increased, and the body weight, the whole heart weight and the left ventricular weight were increased.The myocardial cells were disordered, and the muscle fibers were distorted and occurred.The expression of APN in serum and myocardial tissue was decreased, and the expression of TGF-β1 was increased in the IR group(all P<0.05).Compared with the IR group,the LVEF and LVFS were elevated,FPG and HOMA-IR were decreased, myocardial microstructure was improved, and the expression of APN in serum and myocardial tissue was increased,and the expression of TGF-β1 in myocardial tissue expression was decreased in Ato10 group (P<0.05).The LPVWd and IVSd were decreased,LVEF and LVFS were increased,FPG,FINS and HOMA-IR were decreased,body weight, whole heart weight and left ventricular weight were decreased, myocardial microstructure was further improved, and the expression of APN was increased in serum and myocardial tissue, the expression of TGF-β1 was decreased in Ato20 group, and the improvement effect was more significant in Ato20 group(P<0.05).Conclusion IR can cause ventricular remodeling in mice,and atorvastatin can improve IR-induced ventricular remodeling in a dose-dependent manner,suggesting that its mechanism may be related to elevation of expression of APN.
