摘要:
目的 研究microRNA-451a(miR-451a)在结直肠癌中的表达并观察其对结直肠癌细胞增殖的影响。方法 荧光定量PCR检测miR-451a在24例新鲜的结直肠癌组织样本及4株结直肠癌细胞系HCT116、LoVo、LS174T及SW480中的表达;将miR-451a及NC病毒转染HCT116细胞,荧光定量PCR检测转染后HCT116细胞中miR-451a的表达及细胞增殖、细胞周期相关基因的表达情况;CCK-8及平板克隆形成实验检测HCT116-451a和HCT116-NC细胞的生长增殖情况。结果 与正常结直肠组织比较,miR-451a在结直肠癌组织中表达下调,与正常结直肠细胞系CCD-18Co相比,miR-451a在4株结直肠癌细胞系HCT116、LoVo、LS174T及SW480中表达均下调(P均<0.05)。CCK-8结果显示转染miR-451a的HCT116细胞增殖能力低于NC组,平板克隆形成实验显示与HCT116-NC比较,HCT116-451a组克隆数减少(P<0.05)。与HCT116-NC比较,HCT116-451a组增殖细胞核抗原Ki-67及细胞周期蛋白 D1(Cyclin D1)表达降低,细胞周期素依赖性激酶抑制因子(p21)表达增高(P<0.05)。结论 miR-451a在结直肠癌中低表达,上调miR-451a的表达可抑制结直肠癌细胞HCT116的生长增殖。
Abstract:
Objective To explore the expression of microRNA-451a (miR-451a) in colorectal cancer and its effect on proliferation of colorectal cancer cells. Methods The expression of miR-451a in 24 fresh colorectal cancer tissues and 4 colorectal cancer cell lines(HCT116,LoVo,LS174T,SW480) with different malignant potential was examined by RT PCR.HCT116 cells were transfected with miR-451a or NC virus. The expression of miR-451a and genes related to cell proliferation and cell cycle was detected by RT PCR after transfection. Cell growth and proliferation of HCT116-451a and HCT116-NC were detected by CCK-8 and clone formation.Results Compared with normal colorectal tissues, the expression of miR-451a was down-regulated in 19(a total of 24) colorectal cancer tissues and the expression of mir-451a was also down-regulated in 4 colorectal cancer cell lines including HCT116, LoVo, LS174T and SW480 compared with normal colorectal cell line(CCD-18Co)(P<0.05). CCK-8 results showed that the proliferation ability of HCT116-451a was lower than that of HCT116-NC(P<0.05).Clone formation assay revealed that compared with HCT116-NC,the number of clone was decreased in HCT116-451a(P<0.05).Compared with HCT116-NC, the expression levels of Ki-67 and Cyclin D1 in HCT116-451a group were decreased, while the expression level of p21 was increased(P<0.05). Conclusion The expression of miR-451a was down-regulated in colorectal cancer.Increasing expression of miR-451a can effectively inhibit the proliferation of HCT116 colorectal cancer cells.
