摘要:
目的 探讨miR-29c在矽肺纤维化中的作用。方法 原代培养大鼠肺成纤维细胞,分为阴性对照组(A组)、巨噬细胞培养液刺激组(B组)、miR-29c mimic +巨噬细胞培养液刺激组(C组)和miR-29c无义序列+巨噬细胞培养液刺激组(D组)4组,采用实时荧光定量PCR法检测各组细胞中COL1A1 mRNA表达水平,Western blot法检测COL1A1 蛋白表达水平。结果 与A组比较,B组COL1A1基因与蛋白表达水平均增加(P均<0.05);与B组和D组相比较,C组COL1A1基因与蛋白表达水平明显降低(P均<0.01)。结论 miR-29c可以减少巨噬细胞刺激液诱导的大鼠肺成纤维细胞COL1A1表达,对矽肺纤维化过程有潜在干预作用。
Abstract:
Objective To explore the role of miR-29c in the process of silica lung fibrosis. Methods Primary cultured rats' lung fibroblasts were divided into four groups: negative control group(A group),macrophage stimulating culture fluid group(B group),miR-29c mimic and macrophage stimulating culture fluid group(C group),miR-29c nonsense sequence and macrophage stimulating culture fluid group(D group). RT qPCR and Western blot were used to assay the gene and protein expression level of COL1A1 respectively. Results Compared with group A, the gene and protein expression level of COL1A1 in group B increased significantly(P<0.05). Compared with group B and D,the gene and protein expression level of COL1A1 in group C decreased significantly(P<0.01). Conclusion miR-29c can decrease COL1A1 expression in rats' fibroblast stimulated by macrophage culture fluid, with potential therapeutic effect in silica lung fibrosis process.
