摘要:
目的 探讨阿帕替尼(Axitinib) 减轻磨损颗粒诱导骨溶解的作用及其机制。方法 45只雄性BALB/C小鼠,随机分为对照组、模型组和Axitinib治疗组,每组15只,建立air pouch,模型组和Axitinib治疗组air pouch中注射0.3 mL钛颗粒,对照组air pouch中注射0.3 mL PBS液。Axitinib治疗组7d予以Axitinib(30 mg·kg-1, Bid)灌胃;给药7d 后处死小鼠,对各组小鼠air pouch组织行苏木素-伊红(HE)染色观察air pouch组织炎性反应等情况; 免疫组化染色检测血管内皮生长因子(VEGF)蛋白质表达。 结果 Axitinib治疗组air pouch组织炎症反应明显低于模型组,免疫组化染色结果显示,Axitinib能降低小鼠air pouch组织 VEGF蛋白质表达。结论 Axitinib可以缓解磨损颗粒刺激小鼠air pouch诱导骨溶解中air pouch组织的炎症反应,并且抑制air pouch组织 VEGF 表达的作用。
Abstract:
Objective To explore effects of Axitinib on the expression of VEGF in inflammatory osteolysis induced by wear debris in murine air pouch. Methods 45 male BALB/C mice were randomly divided into control group,model group and Axitinib treatment group,with 15 mice in each group. The air pouch was made. In model group and Axitinib treatment group,0.3 mL titanium granules were injected into the air pouch and 0.3 mL PBS solution was injected into air pouch of control group. Mice in Axitinib treatment group were given Axitinib(30 mg·kg-1,Bid) by gastric perfusion on the 7th day. After 7 days of administration, mice were executed and the inflammatory reaction of air pouch tissue was observed by HE staining. The expression of vascular endothelial growth factor(VEGF) protein was detected by immunohistochemical staining and the mRNA expression was detected by RT-PCR. Results Inflammation of air pouch in Axitinib treatment group was significantly lower than that in the model group. The immunohistochemical staining and RT-PCR results showed that Axitinib could reduce the expression of VEGF protein and mRNA in mice air pouch. Conclusion Axitinib can alleviate the reaction of inflammation in mrine air pouch induced by wear debris,and inhibit the expression of VEGF in air pouch tissue.
