摘要:
目的 探讨不同时期宫内香烟烟雾暴露(cigarette smoke exposure,CSE)对新生大鼠延髓呼吸中枢的损伤作用。方法 SD孕鼠随机分为4组:control组、CSE1~20d组(孕鼠妊娠1~20d进行CSE)、CSE7~20d组(孕鼠妊娠7~20d进行CSE)和CSE14~20d组(孕鼠妊娠14~20d进行CSE),每组8只。与孕鼠相对应的子代新生大鼠作为本研究的实验对象。采用离体延髓脑片灌流技术观察缺氧前后不同时期宫内CSE对新生大鼠离体延髓脑片舌下神经根呼吸节律样放电频率(burst frequency,BF)的影响。结果 各时期宫内CSE对新生大鼠离体延髓脑片舌下神经根基础BF均无明显影响(P均>0.05)。缺氧引起control组BF加快,却引起CSE1~20d和CSE7~20d组BF减慢,而CSE14~20d组BF对缺氧的变化与control组一致(P<0.05);与control组和CSE14~20d组比较,缺氧后CSE1~20d和CSE7~20d组BF均减慢(P均<0.05),CSE1~20d组与CSE7~20d组BF对缺氧的反应差异无统计学意义(P>0.05)。结论 孕第7~14天可能是宫内CSE损伤新生大鼠延髓呼吸中枢的关键时期。
Abstract:
Objective To study the injury of medullary respiratory centers of neonatal rats induced by utero CSE in different pregnant periods. Methods Pregnant SD rats were randomly divided into 4 groups, including control(n=8),CSE1~20d(pregnant rats were exposed to cigarette smoke during gestation days 1~20,n=8), CSE7~20d(exposed to cigarette smoke during gestation days 7~20,n=8) and CSE14~20d (exposed to cigarette smoke during gestation days 14~20,n=8) groups. The neonatal rats were used in the present study. The discharge was recorded from the hypoglossal rootlets of medullary slices from the neonatal rats and the burst frequency(BF) was analyzed. Hypoxia was administered to all the groups of slices to unmask possible indifference of BF responses of rootlets to CSE. Results No significant differences in basal BF were found in 4 groups when the slices were perfused with standard ACSF bubbled with carbogen(P>0.05). The responses of hypoxia was increased in BF in the control and CSE14~20d groups(P<0.05),BF was decreased by hypoxia in the CSE1~20d and CSE7~20d groups(P<0.05). In the CSE1~20d and CSE7~20d groups, compared with the control and CSE14~20d groups, the response of hypoxia decreased in BF(P<0.05). However,the response of hypoxia in BF showed no significant difference between the CSE1~20d and CSE7~20d groups(P>0.05). Conclusion The 7th to 14th day of pregnancy may be the key period of in utero CSE injury to the respiratory center of medulla oblongata in neonatal rats.
