摘要:
目的 观察中介素(intermedin, IMD)对急性心肌梗死大鼠心肌氧化应激及心脏功能的影响。方法 40只SD雄性大鼠随机分为对照组、急性心肌梗死(AMI)组及IMD干预组。干预4周后采用M型超声心动图分别测量左心室收缩末期内径(LVESD)和舒张末期内径(LVEDD);通过HE染色观察梗死缺血区心肌细胞形态变化;测定3组大鼠心肌组织中丙二醛(MDA)水平,超氧化物歧化酶(SOD)活性。结果 与对照组比较,AMI组心肌梗死区出现广泛纤维化、肌纤维断裂、融合,伴有少量淋巴细胞浸润。而IMD干预组可见心肌纤维化及肌纤维断裂、融合现象均有所改善,AMI组LVEDD、LVESD均增大,心肌组织中MDA水平升高,SOD活性下降(P<0.05);与AMI组比较,IMD干预组LVEDD、LVESD均减小,心肌组织中MDA水平降低,SOD活性升高(P<0.05)。结论 IMD能够改善急性心肌梗死大鼠的心脏功能,这种作用可能是通过减少心肌缺血后氧化应激。
Abstract:
Objective To observe the effects of Intermedin(IMD) on oxidative stress and cardiac function in rats with acute myocardial infarction. Methods Forty male SD rats were randomly divided into control group, acute myocardial infarction (AMI) group and IMD intervention group. The left ventricular end-systolic diameter (LVESD) and end-diastolic diameter (LVEDD) were measured by M-mode echocardiography after 4 weeks of intervention; the morphological changes of myocardial cells in infarcted ischemic area were observed by HE staining; the levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) in myocardial tissue were measured in three groups of rats. Results Compared with control group, the myocardial infarction area of AMI group was widely myocardial fibrosis,and the muscle were broken and fused,with a small amount lymphocyte infiltration. The appearance of myocardial fibrosis, muscle fiber fracture and fusion were improved in the IMD intervention group. LVEDD and LVESD were significantly increased in AMI group,MDA contents increased, and SOD activity decreased significantly(P<0.05). Compared with the AMI group,LVEDD and LVESD were significantly decreased in the IMD treatment group, meanwhile the MDA contents was decreased and SOD activity increased significantly(P<0.05). Conclusion IMD can significantly improve the cardiac function of acute myocardial infarction due to ischemia in rats by reducing oxidative stress.
