摘要:
目的 探讨枸杞多糖(lycium barbarum ploysacharides,LBP)对同型半胱氨酸(homocysteine,Hcy)诱导的血管平滑肌细胞(VSMCs)表型转化的抑制作用。方法 使用人脐静脉VSMCs细胞株培养,取4~7代细胞用于实验。实验分为对照组、100μmol·L-1Hcy干预组、Hcy+1g·L-1LBP干预组、Hcy+2g·L-1LBP干预组、Hcy+4g·L-1 LBP干预组共5组。划痕实验测各组VSMCs的迁移情况;光学显微镜观察各组VSMCs的细胞形态;实时定量荧光PCR及Western blot检测各组VSMCs中平滑肌肌动蛋白(SM-actin\SMA)、转凝蛋白(SM22a)、骨桥蛋白(OPN)的mRNA及蛋白的表达。结果 与对照组比较,Hcy组人脐静脉VSMCs增殖迁移率增加(P<0.05),细胞形态变圆,SMA和SM22a mRNA及蛋白表达减少(P<0.05),OPN mRNA及蛋白表达增加(P<0.05)。与Hcy组比较,Hcy+1g·L-1 LBP干预组、Hcy+2g·L-1 LBP干预组人脐静脉VSMCs增殖迁移率减少,细胞形态变的细长,SMA 和SM22a mRNA及蛋白表达增加(P<0.05),而OPN表达减少(P<0.05)。结论 LBP可以抑制Hcy诱导的人脐静脉VSMCs由收缩表型去分化为合成表型,但其效应并不随着LBP浓度的增加而增加。
Abstract:
Objective To investigate the phenotypic transformation of human smooth muscle cells(VSMCs) induced by homocysteine(Hcy) and the intervention of lycium barbarum polysaccharides(LBP). Methods Human umbilical vein VSMCs cells were cultured and the 4~7 generation cells were used for the experiment. The experiment was divided into 5 groups: The VSMCs were divided into 5 groups: control group,100μmol·L-1 Hcy intervention group,Hcy+1g·L-1 LBP intervention group,Hcy+2g·L-1 LBP intervention group and Hcy+4g·L-1 LBP group. The migration of VSMCs in each group was measured by scratch test; the cell morphology of VSMCs in each group was observed by optical microscope;real-time quantitative fluorescence PCR and Western blot were used to detect the expression of mRNA and protein of smooth muscle actin(SM-actin\SMA),SM22a,osteopontin(OPN) in each group of VSMCs. Results The results showed that compared with the control group,the proliferation and migration of VSMCs in Hcy group increased;the cell morphology was round,the expression of SMA and SM22amRNA and protein decreased(P<0.01),and the expression of OPN mRNA and protein increased(P<0.01). Compared with the Hcy group, the proliferation and migration rate of VSMCs in the Hcy+1g·L-1 LBP intervention group and the Hcy+2g·L-1 LBP intervention group decreased,the cell morphologic changes were elongated,SMA and SM22a mRNA and protein expression increased(P<0.05),and the OPN expression decreased(P<0.05). Conclusion LBP can inhibit the differentiation of Hcy induced VSMCs from contractile phenotype to synthetic phenotype,but its effect does not increase with the increase of LBP concentration.
