摘要:
目的 探讨低硒对大鼠心肌组织结构及心功能的影响。方法 将60只纯系断乳SD大鼠随机分为对照组、低硒组(LS组)及补硒组(SS组),每组各20只,对照组喂养标准饲料,LS组大鼠喂养低硒饲料,SS组喂养低硒饲料14周后再给予亚硒酸钠补硒3周,各组喂养17周后,检测大鼠的血硒含量,并采用HE染色观察大鼠心肌组织形态学变化,电镜下观察心肌组织超微结构,采用分类脑钠肽(Triage BNP)快速检测实验检测血浆BNP,心脏超声检测大鼠心功能的变化。结果 LS组、SS组大鼠血清硒水平均低于对照组(P均<0.05),SS组大鼠补硒后血清硒水平上升,高于LS组(P<0.05)。对照组大鼠心肌组织结构正常,LS组大鼠心肌组织结构紊乱,SS组大鼠心肌组织结构仅有轻度异常。与对照组相比,LS组大鼠脑钠肽(BNP)、左心室收缩末期内径(LVESD)、左心室舒张末期内径(LVEDD)均升高(P均<0.05),左心室射血分数(LVEF%)及左室短轴缩短率(LVFS%)均降低(P均<0.05)。SS组大鼠BNP、LVEDD和LVESD水平均低于LS组,LVEF%和LVFS%均高于LS组(P均<0.05)。结论 低硒可导致大鼠心肌组织结构的破坏,进而导致心功能的下降,补硒后可改善大鼠心功能。
Abstract:
Objective To explore the effects of low selenium on myocardial tissue and cardiac function in rats. Methods A total of 60 SD rats were randomly divided into control group,low selenium group and selenium supplement group,with 20 rats in each group. Rats in the control group were fed with standard diet,rats in the low selenium group were fed with low selenium diet and rats in selenium supplement group were fed with low selenium diet for fourteen weeks and then given sodium selenite for three weeks. Seventeen weeks later,blood selenium was detected. HE staining was used to observe the morphological changes of myocardial tissue. Electron microscope was used to observe the myocardial ultrastructure. BNP as well as echocardiography were used to detect the cardiac function in rats. Results The levels of blood selenium in low selenium group decreased significantly compared with the control group,but after selenium supplement they increased significantly(P<0.05). In the control group,the structure of myocardial tissue was normal, but the structure of myocardial tissue in the low selenium group was disordered. There was only mild abnormality in the structure of myocardial tissue in the selenium supplement group. Compared with the control group,BNP,LVESD and LVEDD of rats in the low selenium group was significantly increased,but LVEF% and LVFS% was significantly reduced. The cardiac function was significantly increased after selenium supplement(P<0.05). Conclusion Low selenium can lead to the destruction of myocardial tissue structure,which leads to the decline of cardiac function. Selenium supplement can improve cardiac function in rats.
