摘要:
目的 比较叉头框蛋白A2(forkhead box A2,FOXA2)在结直肠癌细胞系与正常结肠细胞系中的表达差异,并研究FOXA2对结直肠癌LOVO细胞系增殖、迁移以及凋亡的影响。方法 Real-time PCR检测FOXA2 mRNA的表达;在低表达细胞系LOVO细胞中转染过表达FOXA2质粒(FOXA2组)和阴性对照质粒(NC组),以未转染的细胞作为空白对照(CT组),Real-time PCR、Western blot技术验证转染效果;克隆形成实验、划痕实验以及流式细胞术检测过表达FOXA2后结直肠癌LOVO细胞增殖、迁移和凋亡的变化。结果 FOXA2mRNA在4种结直肠癌细胞系中表达均低于正常结肠细胞(P<0.05)。转染FOXA2过表达质粒后,FOXA2组细胞中mRNA和蛋白表达均高于NC组和CT组细胞(P<0.01),NC组细胞中FOXA2 mRNA和蛋白表达与CT组细胞之间差异无统计学意义(P>0.05);与NC组和CT组相比,过表达的FOXA2组LOVO细胞的克隆数和细胞迁移率均降低(P<0.05),NC组和CT组之间的克隆数和细胞迁移率差异无统计学意义(P>0.05);FOXA2组的LOVO细胞凋亡率高于NC组和CT组(P<0.05),NC组细胞凋亡率与CT组差异无统计学意义(P>0.05)。结论 FOXA2在结直肠癌细胞中低表达,过表达FOXA2后会抑制结直肠癌细胞的增殖和迁移,促进癌细胞的凋亡。
Abstract:
Objective The expression levels of Forkhead Box A2 (FOXA2) gene in colorectal cancer cell lines and normal colonic cell lines were compared, and the effects of FOXA2 on the proliferation, migration and apoptosis of colorectal cancer LOVO cell lines were investigated. Methods The FOXA2 plasmids (FOXA2 group) and negative control plasmids(NC group) were transferred into the low expression cell line LOVO cells, and the untransfected cells were used as blank control(CT group). Real-time PCR and Western blot techniques were used to verify the transfection efficiency. The Clone formation assay, scratch test and flow cytometry were used to detect the proliferation, migration and apoptosis of LOVO cells after overexpression of FOXA2. Results The FOXA2 expression levles in four kinds of colorectal cancer cell lines were significantly lower than that in normal colorectal epithelial cells(P all<0.05). FOXA2 mRNA and protein expression were significantly higher than those in NC group and CT group(P all <0.01) after transfection of FOXA2 overexpression plasmids. There were no significant differences in FOXA2 mRNA and protein expression between NC group and CT group (P>0.05). Compared with the control group,the number of cloned cells and migration rate in FOXA2 overexpression group decreased (P<0.05). There was no significant difference in the number of clonal cells and wound healing between the two control groups(NC group and CT group)(P>0.05). The apoptosis rate of LOVO cells in FOXA2 group was significantly higher than that in NC group and CT group. There was no significant difference between NC group and CT group(P>0.05). Conclusion FOXA2 is low expressed in colorectal cancer cells. FOXA2 overexpression can inhibit the proliferation and migration of colorectal cancer cells and promote the apoptosis of cancer cells.
