摘要:
目的 探讨体外弓形虫感染对大鼠肺泡巨噬细胞极化的影响。方法 Diff染色,观察弓形虫在大鼠肺泡巨噬 细胞内的增殖情况。使用荧光定量PCR、Western blot和ELISA等方法,分别从mRNA水平和蛋白质水平检测弓形虫 感染后M1与M2型巨噬细胞相关因子的变化。结果 Diff染色发现,弓形虫感染率在6h至36h无明显变化,在48h感染 率增加(P<0.05)。实时荧光定量PCR结果显示,M1型巨噬细胞相关因子TNF-a、iNOS、CXCL10、CXCL11的mRNA在 短时间内表达上调,而M2型巨噬细胞相关因子TGF-β1、ARG-1mRNA从6h开始升高,在24h达到峰值(P<0.01), CCL17、CCL18分别从24h、12h开始升高,在36h达到峰值(P<0.01)。Western blot结果显示,与对照组相比,弓 形虫感染12h后ARG-1表达开始升高(P<0.05);iNOS无表达。ELISA结果显示,M1型巨噬细胞相关因子TNF-a、IL -12分别在24h、6h上升到峰值(P<0.05);M2型巨噬细胞相关因子,TGF-β1从6h开始升高,在24h达到峰值 (P<0.01),IL-10从24h开始升高,在36h达到顶峰,48h仍高于正常组(P<0.05)。结论 弓形虫感染可以诱导大 鼠巨噬细胞从静息状态极化为M2型,为机体免疫系统与弓形虫的相互作用提供了细胞研究基础。
Abstract:
Objective This study was designed to explore the effect of Toxoplasma gondii infection on rat alveolar macrophage polarization in vitro. Methods Diff staining was used to observe proliferation of Toxoplasma gondii in rat alveolar macrophages. The protein and mRNA levels of target genes were measured by quantitative real-time polymerase chain reaction(RT-qPCR),ELISA and Western blot. Results Toxoplasma gondii infection rate had no obvious change in the early stage but increased significantly at 48h(P<0.05). The RT-qPCR results showed that M1 related factors (iNOS,TNF-a,CXCL10,CXCL11) increased during a short period of time and had no obvious change compared with control at other times. M2 related factors (ARG-1,TGF-β1) increased from 6h and reached peak at 24h. M2 related factors(CCL17,CCL18) increased from 24h and 6h and reached peak at 36h without no significant decrease trend. Western blot results showed that ARG-1 began to increase from 12h,but iNOS was not detected. ELISA results also showed that TNF-a(M1 related factors) and IL-12 had a significant rise at 24h and 6h(P<0.05). M2 related factors TGF-β1 increased from 6h and reached peak at 24h,IL-10 increased from 24h and reached peak at 36h. And no significant decrease was observed at the later time points. Conclusion Toxoplasma gondii infection can induce alveolar macrophage from resting stage to M2. The study contributes to further elucidate the interacting between Toxoplasma gondii and host' immune system.
