摘要:
目的 观察苦豆碱对异丙肾上腺素所致大鼠心肌肥厚的保护作用。方法 雄性Sprague-Dawley大鼠,随机分 为5组:正常对照组、模型组及10、20、40mg·kg-1苦豆碱组,连续灌胃给药14 d。于第8天皮下注射异丙 肾上腺素(ISO,5mg·kg-1,连续 7 d) 建立大鼠心肌肥厚模型。观察苦豆碱对心肌肥厚大鼠左心室内压最大上 升速率(+LVdp/dtmax)、左室收缩压(LVSP)、左心室内压最大下降速率(-LVdp/dtmax)、平均动脉压、心率 、心肌肥厚指数、心肌组织形态结构的影响;分离血清,检测苦豆碱对心肌肥厚大鼠心肌肌钙蛋白(cTnⅠ)及丙 二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)等的影响。结果 与模型组比较,20和40 mg·kg-1苦豆碱可降低大鼠全心湿质量、左心室湿质量、全心湿质量/体质量比与左心室湿质量/体质量比 ,升高+LVdp/dtmax、-LVdp/dtmax,降低LVEDP(P<0.05);苦豆碱可减轻大鼠心肌细胞肥大、纤维化、间质水 肿和炎细胞浸润等ISO导致的心肌组织病理学结构的异常改变;同时降低cTnⅠ 水平(P<0.01)。此外,20mg ·kg-1和40mg·kg-1苦豆碱能提高血清中SOD和GSH-PX活性,并降低MDA的含量(P<0.05)。结论 苦 豆碱对ISO所致大鼠心肌肥厚具有保护作用,其作用机制可能与提高大鼠的抗氧化能力有关。
Abstract:
Objective To investigate the protective effect of aloperine on isoproterenol-induced cardiac hypertrophy in rats and its mechanisms.Methods Male Sprague Dawley rats were given aloperine(10,20,40 mg·kg-1) orally for 14 days. Myocardial hypertrophy model was induced in rats by subcutaneous injection of isoproterenol(5 mg·kg-1) at the 8th day for 1 week. The haemodynamic parameters,the body weight,myocardial hypertrophy indexes and histomorphology were checked to evaluate the effect of aolperine on isoproterenol-induced myocardial hypertrophy in rats. In addition,levels of cardiac troponinsⅠ,superoxide dismutase,malondialdehyde and glutathione peroxidase in serum were measured. Results Compared with model group,oral administration of aloperine considerably decreased LVEDP and increased LVdp/dtmax on myocardial hypertrophy rats.Aloperine treatment significantly decreased the myocaridial hypertrophy indexes and attenuated myocardial necrosis,interstitial edema and inflammatory cell infiltration. Moreover ,aloperine decreased the level of cardiac troponinsⅠand increased antioxidant enzyme activity to different degrees and enhanced the activity of superoxide. Conclusion These results demonstrate that aloperine exerts protective effects on isoproterenol-induced myocardial hypertrophy in rats, which is likely to be mediated through increasing antioxidant enzyme activities and normalization the balance of oxidation and antioxidation.
