摘要:
目的 探索吗啡和舒芬太尼对新生SD大鼠原代心肌细胞缺氧/复氧损伤模型细胞增殖情况与缝隙连接蛋白43(Cx43)及其磷酸化的影响。方法 原代心肌细胞随机分为正常对照组(C组)、缺氧/复氧组(I/R组)、吗啡组(MF组)和舒芬太尼组(SF组),MF组和SF组分别用吗啡(0.3μmol·L-1)和舒芬太尼(0.0003μmol·L-1)分别预处理16min后,除去药物,培养20 min; I/R组、MF组、SF组均缺氧2h、复氧1h。用CCK-8试剂盒检测各组的细胞增殖情况; 激光共聚焦显微镜观察各组心肌细胞中Cx43的分布与表达; 用Western blot法检测Cx43总蛋白及磷酸化Cx43(pCx43)的表达。结果 与C组相比,I/R组、MF组、SF组细胞增殖减少(P<0.01); 与I/R组相比,MF组、SF组细胞增殖均增多(P<0.01); MF组细胞增殖多于SF组(P<0.05)。与C组相比,I/R组、SF组、MF组Cx43荧光分布散乱以及强度减弱、Cx43和pCx43表达量减少(P<0.01); 与I/R组相比,MF组和SF组Cx43荧光分布较连续、荧光强度较高(P<0.01),Cx43总蛋白表达量以及磷酸化水平较高(P<0.01); MF组与SF组比较,Cx43荧光分布较整齐、荧光强度较高(P<0.01),磷酸化Cx43表达量较高(P<0.01)。结论 舒芬太尼和吗啡对缺氧/复氧损伤大鼠心肌细胞均有保护作用,其作用可能与心肌细胞Cx43表达及其磷酸化水平有关,吗啡的心肌细胞保护作用及其Cx43磷酸化作用强于舒芬太尼。
Abstract:
Objective To explore effect of morphine and sufentanil on cell proliferation, connexin 43(Cx43)and phosphorylated Cx43 in primary myocardial hypoxia-reperfusion relational model of neonatal SD rats. Methods Primary cardiomyocytes were randomly divided into normal control group(group C), hypoxia / reoxygenation group(I / R group), morphine group(MF group)and sufentanil group(SF group). Rats of MF morphine group(0.3μmol·L-1)and SF Sufentanil group(0.0003μmol·L-1)were pretreated for 16 minutes, removed drugs and trained 20 min. Rats of I / R group, MF group, SF group were performed hypoxia 2h, reoxygenation 1h. CCK-8 kit was used to observe cell proliferation of each group while laser scanning confocal microscope was used to observe the distribution of myocardial cells and expression of Cx43. Cx43 expression and phosphorylation of total protein Cx43(pCx43)were detected by Western blotting method. Results Compared with rats of the C group, cell proliferation of rats in I/Rgroup,MF group,SF group decreased significantly(P<0.01). Compared with rats of I/R group, cell proliferationof rats in MF group, SF group were significantly increased(P<0.01). Compared with rats of MF group, cell proliferation rats in SF group got down significantly(P<0.05). Compared with rats of the C group, Cx43 fluorescence intensity distribution in I/R group,SF group, MF group scattered and weakened and expression of Cx43 and pCx43 significantly reduced(P<0.01). Compared with rats of I/R group, Cx43 fluorescence distribution in MF group and SF group was more continuous and higher fluorescence intensity(P<0.01), higher total protein Cx43 expression and phosphorylation levels(P<0.01). MF phosphorylation of Cx43 expression group was significantly higher than that of the SF group(P<0.01). Cx43 fluorescence distribution and intensity in MF group was neater and higher than that in SF group(P<0.01). Conclusion Sufentanil and morphine have a protective effect on hypoxia / reoxygenation injury in myocardial cells. Its effect may be related to Cx43 expression and phosphorylation of myocardial cells. Effect of morphine on myocardial protection and improve their level of phosphorylation of Cx43 was stronger than that of sufentanil.
