摘要:
目的探讨高血压对心脏的影响及可能的分子机制。方法测量不同周龄Wistar-kyoto大鼠(WKY)和自发性高血压大鼠(SHR)血压值及心脏重量,显微镜测微尺观测不同周龄SHR心脏构型,透射电子显微镜观察心肌细胞的改变,免疫组化和Western-blotting方法观察p38MAPK和磷酸化p38MAPK在高血压心脏重构中表达情况。结果8周龄开始,SHR组血压逐渐升高,并随着周龄的增加而增加(P<0.05),SHR组动脉收缩压明显高于同周龄WKY组(P<0.05)。16、24周龄SHR大鼠的心脏/体重比值明显高于4周龄SHR,同时明显高于同周龄WKY大鼠(P<0.05)。16、24周SHR组左心室壁厚度明显高于4周龄SHR也明显高于16、24周WKY大鼠(P<0.05)。SHR组,磷酸化p38在左心室肌表达随周龄的增加逐渐增加,在SHR24表达明显高于SHR4周龄组(P<0.05)。磷酸化p38在SHR组左心室表达明显高于同周龄WKY大鼠(P<0.05)。结论长期高血压可导致心脏重构,心脏重构改变随周龄增加逐渐加重,p38MAPK信号通路磷酸化可能参与了高血压心脏重构。
Abstract:
Objective To explore the effects of hypertension on the heart and its possible underlying molecular mechanism. Methods Blood pressure and heart weight of Wistar-kyoko rats(WKY)and spontaneously hypertensive rats(SHR)were measured on different week. Microscope Graticules and transmission electron microscope were used to detect cardiac geometric alteration of spontaneously hypertension rats(SHR)and cytological changes of myocardial cell following hypertension progress. Immunohistochemical staining and Western-blotting were applied to observe the expression of p38MAPK and phosphorylated p38MAPK in myocardial cell of SHR. Results From 8 weeks of age, the blood pressure of the SHR groups gradually increased. With the rats grew up, the systolic blood pressure increased(P<0.05). The blood pressure of the SHR groups was significantly higher than that of age-mathched Wistar kyota(WKY)rats(P<0.05). The ratio of heart weight to body weight of SHR16 and SHR24 was significantly higher than that of SHR4, WKY16 and WKY20, respectively(P<0.05). The left ventricular wall thickness of SHR16 and SHR24 was significantly higher than that of SHR4,WKY16 and WKY20, respectively(P<0.05). The increase expression of phosphorylated p38 in the left ventricle was found with age increase of SHR. The expression of phosphorylated p38 in SHR24 was significantly higher than that in SHR4(P<0.05).The expression of phosphorylated p38 in the left ventricle of the SHR groups was significantly higher than that of age matched WKY rats controls(P<0.05). Conclusion Prolonged high blood pressure can induce cardiac remodeling. The cardiac remodeling was aggravated by gradually increase of hypertenison. The p38 MAPK signal pathway phosphorylation and activation might be involved in hypertensive cardiac remodeling.
