摘要:
目的观察硫酸铍(BeSO4)对BALB/C小鼠脾脏淋巴细胞增殖及T细胞亚群的影响。方法BALB/C小鼠随机分为对照组和BeSO4染毒低(2.5mg·kg-1)、中(5.0mg·kg-1)、高(10.0mg·kg-1)剂量组。每周称量小鼠体重一次,染毒结束后小鼠脱颈椎处死,无菌取出脾脏并称重。MTT法检测小鼠脾脏淋巴细胞增殖,流式细胞术测定T淋巴细胞亚群。结果BeSO4染毒5周后,低、中、高剂量组BALB/C小鼠体重均下降,其中中、高剂量组与对照组比较差异有统计学意义(P<0.05);低、中、高剂量BeSO4染毒组脾脏重量及脾脏系数均高于对照组(P<0.05);经BeSO4染毒后小鼠脾脏淋巴细胞增殖能力增强,低、中、高剂量组与对照组之间差异有统计学意义(P<0.05);三种剂量BeSO4染毒均可明显增加小鼠脾脏CD4+T细胞数量及CD4+T细胞与CD8+T细胞的比值(P<0.05)。结论BeSO4可致BALB/C小鼠细胞免疫功能紊乱。
Abstract:
Objective To observe the Beryllium sulfate(BeSO4)after the intervention analysis of BeSO4 in BALB/C mice spleen lymphocyte proliferation and the lymphocyte subsets. Methods Mice were randomly divided into control group and low BeSO4 infected(2.5 mg·kg-1), medium(5.0 mg·kg-1), high dose group(10.0 mg·kg-1). The mice spleen lymphocyte proliferation and T lymphocyte subsets were determined by MTT method and flow cytometry, respectively. Results BeSO4 administration after 5 weeks, low, medium and high dose group of BALB/C mice body weight decreased while the middle, high dose group had significant difference compared with control group(P﹤0.05). Low, medium and high dose spleen weight, spleen coefficient BeSO4 group were significantly higher than those of the control group(P﹤0.05). After BeSO4 contaminated mice spleen lymphocyte proliferation ability enhancement, low, medium and high dose group and control group showed significant difference(P﹤0.05). Three doses of BeSO4 could significantly increase mice spleen CD4+T cells and CD4+T cells and the ratio of CD8+T cells(P﹤0.05). Conclution BeSO4 can cause BALB/C mice cellular immune dysfunction.
