摘要:
目的研究老瓜头生物总碱的体外抗氧化活性。方法用紫外分光光度计测定老瓜头生物总碱与K3Fe(CN)6作用后吸光度的变化,反应还原力的大小,用Elisa测定老瓜头生物总碱对双氧水诱导的羟自由基的清除能力,用紫外分光光度计测定老瓜头生物总碱对双氧水诱导小鼠红细胞溶血的保护作用,用Elisa测定老瓜头生物总碱对双氧水诱导的肝脂质过氧化产物MDA的清除能力,观察老瓜头生物总碱的抗氧化活性。结果老瓜头生物总碱0.2、0.4、0.8、1.6、3.2mg·mL-1浓度组与相应浓度维生素C相比,还原力均较弱(P<0.01);在0.4、0.8、1.6、3.2mg·mL-1浓度组老瓜头生物总碱与维生素C相比,对羟自由基的清除能力强(P<0.01);老瓜头生物总碱浓度为0.2、0.4mg·mL-1时对红细胞膜的抑制作用与维生素C相比有统计学差异(P<0.05);质量浓度为0.8、1.6mg·mL-1老瓜头生物总碱对小鼠肝匀浆自发氧化生成MDA有抑制作用,其效应强于相应浓度维生素C(P<0.01)。结论老瓜头生物总碱还原力较相应浓度维生素C弱但有较强的清除自由基的能力。
Abstract:
Objective To explore the antioxidant activity of total alkaloid of Cynanchum Komarovii AI.Icjiniski in vitro. Methods Antioxidant activity of total Alkaloid of Cynanchum Komarovii AI.Icjiniski were analyzed in different systems including reducing power, hydroxyl radical scavenging, H2O2-caused hemolysis and lipid peroxidation assay. Vitamin C(Vc)were used as the standard antioxidants. Results Total Alkaloid of Cynanchum Komarovii AI.Icjiniski could scavenge hydroxyl radical specifically and inhibit MDA generation caused by hydroxyl radical,antagonize hemolysis induced by H2O2,but it did not affect reducing power significantly. Conclusion Total Alkaloid of Cynanchum Komarovii AI.Icjiniski could scavenge hydroxyl radical specifically in vitro but reducing power was weak.
