摘要:
目的探讨胰岛素样生长因子结合蛋白4(IGFBP4)在左旋精氨酸诱导的小鼠急性坏死性胰腺炎中的表达和意义。方法利用腹腔注射左旋精氨酸诱导小鼠急性坏死性胰腺炎模型,以生理盐水腹腔注射作为对照组。应用酶联免疫吸附实验(ELISA)检测血清中的IGFBP4的含量;免疫组化染色及WesternBlot检测IGFBP4在胰腺组织中的分布及表达。结果小鼠模型组用左旋精氨酸造模后3d,血清中的IGFBP4含量(1.15±0.35)μg·mL-1明显高于生理盐水对照组(0.39±0.03)μg·mL-1(P<0.05)。免疫组化染色显示:在模型组中可见部分腺泡细胞中IGFBP4染色呈强阳性,且伴随胰腺损伤坏死的严重程度,IGFBP4表达阳性的腺泡细胞越多。Western-blot检测显示,与对照组相比较,模型组中的IGFBP4蛋白表达明显升高。结论在小鼠坏死性胰腺炎中,IGFBP4表达明显上调,且主要表达于胰腺坏死区域中的残存腺泡细胞内,提示IGFBP4可能参与坏死性急性胰腺炎的发病过程。
Abstract:
Objective To investigate the expression and function of the insulin-like growth factor binding protein 4(IGFBP4)in the L-arginine in mice induced by acute necrotizing pancreatitis. Methods The model of the mice acute necrotizing pancreatitis was induced by intrapertoneal injection of L-arginine. The expression of IGFBP4 in serum was detected by ELISA. The protein expression of IGFBP4 in the pancreatic tissue were detected by immunohistochemical staining and Western-blot. Results After modeling three days, the serum levels of IGFBP4 in model group(1.15±0.35)μg·mL-1 was significantly higher than those in saline control group(0.39±0.03)μg·mL-1(P<0.05). Immunohistochemical staining showed that the IGFBP4 was strongly positive stained in the areas of injury acinar cells of model group, and accompanied with the necrosis of pancreatic injury severity. Western-blot analysis showed that the IGFBP4 protein expression of model group was significantly increased than control group. Conclusion In mice with necrotizing pancreatitis, IGFBP4 expression significantly up-regulated and mainly expressed in the remnants of acinar cells in the area of pancreatic necrosis. It suggested that the IGFBP4 might be involved in the pathogenesis of necrotizing acute pancreatitis.
