摘要:
目的 研究羟基类固醇硫酸基转移酶SULT2B1对小鼠H22肝癌细胞增殖能力的影响及作用机制。方法 用不同MOI值(50,100)的SULT2B1慢病毒干扰载体感染H22细胞,CCK-8法检测干扰SULT2B1对H22细胞增殖能力的影响,采用Real-time PCR和Western blot检测增殖相关基因BCL2和MYC的mRNA及蛋白表达水平。结果 与对照组相比,干扰SULT2B1显著抑制H22细胞在不同时间点的增殖能力(MOI=100)(P<0.05)。不同MOI值(50,100)感染显著抑制H22细胞中SULT2B1基因的表达,并降低BCL2与MYC的mRNA及蛋白水平(P<0.05),但不同MOI值间各项指标的差异不明显(P>0.05)。结论 干扰SULT2B1可抑制H22细胞的体外增殖能力,其作用主要与下调BCL2和MYC基因的表达水平有关。
