摘要:
目的 探讨肝癌缺失基因1(delete in liver cancer 1,DLC-1)及基质金属蛋白酶-9(MMP-9)在胃癌黏膜表达的意义。方法 采用免疫组化学技术检测DLC-1与MMP-9在43例胃癌黏膜中和20例正常胃黏膜中DLC-1与MMP-9蛋白表达情况,实时荧光定量逆转录聚合酶链反应法检测30例胃癌黏膜和15例胃正常黏膜中DLC-1与MMP-9基因表达情况。结果 DLC-1蛋白阳性表达在胃癌黏膜中(44.2%),明显低于正常胃黏膜(100%),P<0.05; MMP-9蛋白阳性表达在胃癌黏膜中(72.1%),显著高于胃正常黏膜的阳性表达(20%),P<0.05; DLC-1蛋白表达减弱和MMP-9蛋白表达增强与肿瘤的侵袭程度、淋巴结转移、TNM分期显著相关(P<0.05); 胃癌黏膜与正常胃黏膜中DLC-1mRNA相对表达量分别为(0.26±0.09)、(0.77±0.36)t=7.4283,P<0.05,MMP-9mRNA表达量分别为(9.87±5.69)、(4.23±2.01)t=3.7035,P<0.05; 胃癌黏膜中DLC-1和MMP-9蛋白的表达及基因表达均呈负相关。结论 低表达DLC-1和高表达MMP-9可能与胃癌的发生、浸润和转移密切相关,在胃癌发生发展中存在DLC-1负调控MMP-9的作用。
Abstract:
Objective To investigate the expression of DLC-1 and MMP-9 in gastric cancer tissues, and to analyze their relationship as well as clinical significance. Methods Immunohistochemistry was used to examine the expression of DLC-1 and MMP-9 proteins in gastric carcinoma(n=43)and normal gastric tissues(n=20). The relationship with clinico-pathological characteristics of gastric carcinoma was analyzed. Real-time polymerase chain reaction(RT-PCR)was used to detect mRNA of DLC-1 and MMP-9 in gastric carcinoma(n=30)and normal gastric tissues(n=15). Results(1)The positive expression rate of DLC-1 protein in gastric cancer tissues(44.2%)was significantly lower than that in normal tissue(100%)(P<0.05).The positive expression rate of MMP-9 in gastric cancer tissues(72.1%)was significantly higher than that in normal gastric tissue(20%)(P<0.05).(2)The low expression of DLC-1 and the high expression of MMP-9 had relation with the depth of infiltration, lymph node metastasis and TNM staging of gastric carcinoma(P<0.05). It is a negative correlation between DLC-1 and MMP-9 in the protein expression level and in gene expression level. Conclusion DLC-1 and MMP-9 may be associated with the occurrence of gastric cancer invasion and metastasis, and they may be play a negative coordinating role in the development of gastric cancer.
