摘要:
目的 探讨细粒棘球蚴重组HSP70蛋白(rEgHSP70)免疫小鼠对原头蚴攻击感染的保护性效果。方法 选取雌性6周龄ICR小鼠随机分为两组:实验组和对照组(每组10只),实验组接种rEgHSP70 3次,每次间隔两周,对照组注射等量的PBS。两组小鼠在末次免疫后2周用1500只活细粒棘球蚴进行腹腔攻击感染,攻击感染30周后剖杀小鼠, 无菌取脾组织,并记录每只小鼠腹腔内包囊数量和直径,评价免疫保护力。用流式细胞仪检测脾组织中 CD4+T细胞和 CD8+T 细胞,计算两者比值。结果 细粒棘球蚴重组HSP70蛋白接种对原头蚴攻击感染的免疫保护力为44.20%,rEgHSP70免疫组小鼠脾CD4+T亚群(0.52±0.082)高于对照组小鼠(0.44±0.07),P<0.05,rEgHSP70免疫组小鼠脾淋巴细胞 CD4+/ CD8+T亚群比值(3.35±0.02)高于对照组(2.00±0.77),P<0.05。结论 细粒棘球蚴重组HSP70可诱导小鼠产生44.20%免疫保护力,具有一定的疫苗潜力。
Abstract:
Objective To identify vaccine candidate character of recombinant HSP70 againsthydatidosis,through detecting the immunoprotection. Methods Six weeks old female ICR mice were randomized into two group of 10 mice in each. The mice in test group were immunized three times with an interval of two weeks and those in control group with equal PBS. The mice in two group were challenged by with 1500 Eg protoscoleces intraperitoneally on tenth week after immunization. The mice were sacrificed 30 weeks after infection and Spleens were taken for detecting CD4+T cells and CD8+T cells and ratiocalculated. Results Mice immunized with rEg. HSP70 produced effective immunity protection(reductions in cyst load are 44.2%). Compared to control groups, the CD4+T cells(0.5223±0.0817)and CD4+/CD8+ratio(3.349±0.0160)were significantly higher in the rEg. HSP70 group(both P<0.05). Conclusion Recombinant HSP70 vaccine of Echinococcus granulosus shows partial immune protection. Therefore it can be used as a vaccine candidate in the future.
