摘要:
目的 观察邻苯二甲酸二(2-乙基已基)酯(di-(2-ethylhexyl)phthalate,DEHP)低剂量暴露对初断乳小鼠生长发育的影响。方法 利用亚慢性毒性试验的方法,采用经口灌胃的染毒方式,将36只初断乳昆明种小鼠随机分为对照(玉米油)组和两个DEHP实验组(100mg·kg-1组、500mg·kg-1组),每周染毒5d,休息2d,每天早上8:30开始灌胃染毒,重复染毒11周。每组12只,雌雄各半。记录比较小鼠每周的体重、摄食量,11周后,水合氯醛腹腔麻醉,心尖采血处死,取小鼠脏器称重并计算脏器系数; 取脾脏的1/2进行研磨制备细胞悬液,应用流式细胞仪测定脾脏CD4+T、CD8+T的个体百分量。结果 各组小鼠在染毒期间,体重及其体重增长量与对照组比较差异无统计学意义(P>0.05),各组肝脏、脾脏、胸腺、双肾脏、生殖器周围白色脂肪、腓肠肌脏器系数与对照组比较差异无统计学意义(P>0.05)。DEHP染毒组小鼠脾脏CD4+T、CD8+T百分比,与对照组比较差异均无统计学意义(P>0.05); DEHP染毒组小鼠脾脏CD4+/CD8+比值,与对照组比较差异具有统计学意义(P<0.05)。结论 DEHP低剂量暴露可能对初断乳小鼠细胞免疫水平具有一定抑制作用。
Abstract:
Objective To observe the influence of low dose of ester di-(2-ethylhexyl)phthalate on growth and development in early ablactation mice. Methods Using the chronic toxicity test method, 36 kunming mice who were early ablactation were randomLy divided into control group(corn oil group)and two DEHP experimental group(100 mg·kg-1 group, 500 mg·kg-1 group), 12 in each group, half male and half female. Body weight and food consumption were recorded and compared weekly in mice, splenic cell suspension was prepared and CD4+, CD8+ were measured by flow cell analyzer. Results The body weight and body weight gain of the mice in each group in the exposure period showed no signifcant difference among groups(P>0.05). Spleen lymphocytes CD4+, CD8+ percentage in DEHP infected mice, comparing with corn oil group, showed no significant difference(P>0.05). Spleen lymphocytes CD4+ / CD8+ ratio in DEHP infected mice, compared with corn oil group showed statistically significant difference(P<0.05). Conclusion DEHP low dose exposure may has a certain inhibition to immune function development in early ablactation mice.
