摘要:
目的 对结核分枝杆菌H37Rv(宁夏分离株NXT1024)MPT64基因进行生物信息学预测和分析,通过与结核杆菌标准株序列的比对,观察其变异情况。方法 提取结核分枝杆菌H37Rv(宁夏分离株NXT1024)基因组,以该基因组为模板,采用聚合酶链反应(PCR),从中扩增出MPT64基因,并测序,测序结果利用DNA star软件初步预测出该蛋白的分子特征、抗原性和二级结构等生物信息学信息。结果 MPT64基因全长为687bp,BLAST结果显示基因序列与GeneBank公布的标准株的基因序列的核苷酸同源性为93.6%,对该序列编码蛋白质的构象测试发现,该MPT64编码蛋白含有多个β转角结构,及多个明显的具有抗原活性的结构域和免疫优势辅助性T 淋巴细胞抗原位点。结论 以结核分枝杆菌宁夏分离株为模板成功扩增MPT64基因,该株与标准株的核苷酸同源性为93.6%。通过生物信息学分析获得了该蛋白的蛋白构象、抗原活性的结构域及T细胞表位等相关的生物学信息特征。
Abstract:
Objective To amplify and analyze the MPT64 gene related to the protein secretion of Mycobacterium tuberculosis(Ningxia isolates)and so as to search it's role as new drug target or to diagnose Tuberculosis. Methods The MPT64 gene extracted from the genomic DNA of Mycobacterium tuberculosis H37Rv strain(Ningxia isolates)was amplified by PCR and identification with nucleotide sequencing analysis. Then its genome sequence and encoding proteins was analyzed by DNAstar. Results The MPT64 fragment was composed of 687base pairs and the nucleotide homology with type strain on the GenBank was 93.6%. Conclusion The target gene had been amplified which provided the basis for further researching on its biological function.
