摘要:
目的 通过高脂高糖喂养方法建立一种与人类妊娠糖尿病相似的妊娠糖尿病小鼠模型。方法 将60只C57BL/6J孕鼠随机分为普通饮食孕鼠组和高脂高糖孕鼠组,确定受孕成功之日起,普通饮食孕鼠组给予普通饲料喂养,高脂高糖孕鼠组给予高脂高糖饲料喂养。分别于妊娠0、10、18d测空腹血糖与体重; 于妊娠18d酶联免疫吸附法测定空腹胰岛素并计算胰岛素抵抗指数; 比较孕鼠成模情况。结果 高脂高糖喂养C57BL/6J孕鼠建立妊娠糖尿病小鼠模型的成模率为53.3%; 与正常对照组比较,妊娠第10、18天GDM模型组小鼠血糖水平升高,体质量增加(P<0.05); 妊娠第18天GDM模型组空腹胰岛素及胰岛素抵抗指数均高于正常对照组(P<0.05)。结论 高脂高糖喂养C57BL/6J孕鼠建立妊娠糖尿病小鼠模型自妊娠中期血糖升高,至妊娠晚期仍保持高血糖水平,妊娠晚期存在明显的胰岛素抵抗,这些特征接近人类妊娠糖尿病的发病特点。
Abstract:
Objective To establish a gestational diabetes mouse model by feeding high fat and high sugar diet. Methods 60 C57BL/6J pregnant mice were randomly divided into normal control group and high fat and high sugar feeding group. From the date of pregnancy success, the normal control group was given normal diet while high fat and high sugar feeding group were given a high fat and high sugar feeding. On the pregnancy 0,10, 18 day, the fasting plasma glucose and the body weight were measured. On the pregnancy 18 day, the insulin levels and the insulin resistance index were measured. The modeling rate of the high fat and high sugar feeding GDM model were computed. Results The modeling rate of the high fat and high sugar feeding GDM model was 53.3%. The fasting plasma glucose and the body weigh of high fat and sugar feeding GDM group increased on the pregnancy 10 and 18 day.The insulin levels and the insulin resistance index of high fat and sugar feeding GDM group was also higher than that in control group. Conclusion The high fat and high sugar feeding C57BL/6J pregnant mice to establish a mouse model of gestational diabetes remains high blood glucose level from the second trimester to the third trimester of pregnancy. The third trimester of pregnancy has obvious insulin resistance. These characteristics are similar to human being of gestational diabetes.
