摘要:
目的 研究骨髓间充质细胞向神经样细胞诱导分化的能力,探讨利用其治疗神经损伤疾病的可行性。方法 采用直接分离培养法从人骨髓中分离培养骨髓间充质干细胞,流式细胞术对其细胞特性进行鉴定,化学诱导法诱导其向神经样细胞分化,免疫组化技术验证诱导细胞特性。结果 成功获得骨髓间充质干细胞,免疫表型鉴定结果为CD73、CD90、CD105表达阳性,CD14、CD34、CD45表达阴性; 免疫组化染色显示诱导分化的细胞表达神经前体细胞标志物Nestin、GFAP和成熟神经元标志物NSE等。结论 骨髓间充质细胞在特定诱导因子作用下能向神经细胞表型转化,并能稳定表达神经细胞因子,可以作为神经损伤疾病组织工程的种子细胞来源。
Abstract:
Objective To explore the neural differentiation ability of bone marrow Mesenchymal stem cells(BMSCs). Methods BMSCs were isolated and cultured directly from bone marrow. The cell surface marker were analyzed with Flow Cytomytry. BMSCs differentiation into neural cells were induced by chemistry factor. Immunohistochemical was used to identify cell types. Results The BMSCs were got successfully. The cells positively expressed CD73,CD90,CD105, and negatively expressed CD14,CD34,CD45. Immunohistochemical staining displayed the differentiation cells expressed neural precursor cells markers Nestin, GFAP and mature neurons markers NSE. Conclusion BMSCs could be induced to neural cells. It could be used as seed cells for neural injured disease clinical application.
