摘要:
目的 观察与比较黄芪甲苷和黄芪注射液对大鼠脑出血灶周围神经元Caspase-3阳性细胞数和Caspase-3 mRNA表达的影响。方法 将128只雄性SD大鼠随机分为正常组、模型组、黄芪注射液组、黄芪甲苷组。用药组于造模型后0h腹腔注射用药,继之每隔24h注射一次; 正常组和模型组注射相应量的生理盐水。分别在1、2、3、7d通过免疫组化和RT-PCR方法来观察Caspase-3阳性细胞数和Caspase-3 mRNA表达以及二者的相关性。结果 与正常组相比,模型组、黄芪注射液组、黄芪甲苷组Caspase-3阳性细胞数和Caspase-3 mRNA表达均明显增加(P<0.05); 黄芪甲苷和黄芪注射液的2、3、7d组分别与同时间点模型组比较,Caspase-3阳性细胞数和Caspase-3 mRNA表达均明显降低(P<0.05); 黄芪甲苷和黄芪注射液的7d组分别与各自的3d组比较,Caspase-3阳性细胞数和Caspase-3 mRNA表达均明显降低(P<0.05); 与黄芪注射液3、7d组相比,黄芪甲苷同时间点组的Caspase-3阳性细胞数和Caspase-3 mRNA表达均明显升高(P<0.05); Caspase-3 mRNA与Caspase-3阳性细胞数呈正相关(r=0.97,P<0.05)。结论 黄芪甲苷和黄芪注射液可能是通过降低Caspase-3 mRNA转录,减少Caspase-3蛋白表达,抑制大鼠脑出血灶周围神经元凋亡; 随着用药时间的延长,对出血灶周围神经元凋亡的抑制作用更强; 黄芪甲苷对大鼠脑出血灶周围神经元凋亡的抑制作用不如黄芪注射液。
Abstract:
Objective To observe and compare the effect of Astragaloside IV and Astragalus injection on Caspase-3 positive cells and Caspase-3 mRNA expression of neurons around intracerebral hemorrhage in rats. Methods 128 male SD rats were randomly divided into normal group, model group, Astragalus injection group and Astragaloside IV group. Treatment groups were intraperitoneally injected with Astragaloside IV and Astragalus on 0 h after establishing model, then every 24 h injection was conducted. The normal group and model group were injected saline. Caspase-3 positive cells and Caspase-3 mRNA expression as well as the correlation between the both were observed with by immunohistochemistry and RT-PCR method at 1st d,2nd d,3rd d,7th d respectively. Results Compared with the normal group, Caspase-3 positive cells and Caspase-3 mRNA expression of Astragalus model group, injection groups and Astragaloside IV groups were significantly increased(P<0.05). Compared with the model group at the same time, Caspase-3 positive cells and Caspase-3 mRNA expression in Astragaloside IV and Astragalus injection groupat 2nd d, 3rd d, 7th d were significantly lowered(P<0.05). Compared with the 3rd d treatment group, Caspase-3 positive cells and Caspase-3 mRNA expression in Astragaloside IV and Astragalus injection 7th d group were significantly lowered(P<0.05). Compared with Astragalus injection 3rd d group, 7th d group, Caspase-3 positive cells and Caspase-3 mRNA expression of Astragaloside IV group at the same time were significantly increased(P<0.05). Caspase-3 mRNA and Caspase-3 positive cells showed significantly positively correlated(r=0.97,P<0.05). Conclusion The Astragaloside and Astragalus injection may inhibited neurons apoptosis of brain hemorrhage in rats through reduction of caspase-3 mRNA transcription and the Caspase-3 protein expression. With the extension of the medication time, the inhibition of neurons apoptotic around rats intracerebral hemorrhage are more obvious. Astragaloside IV inhibition of neurons apoptotic around rats intracerebral hemorrhage is worse than the Astragalus injection.
