摘要:
目的 观察不同剂量枸杞叶和枸杞多糖对不同月龄快速老化模型小鼠(SAMP8)老化征象和学习记忆能力的影响。方法 选用 3、5、7 月龄SAMP8,以同龄正常老化小鼠(SAMR1)为正常对照。SAMP8随机分为给药组和对照组,给药组分为枸杞叶全汁高(枸杞叶1.0g·kg-1)、低剂量组(枸杞叶0.5g·kg-1)和枸杞多糖高(0.1g·kg-1)、低剂量组(0.05g·kg-1); 对照组分为安慰剂对照组和空白对照组。给药组灌胃服用高、低剂量枸杞叶全汁或枸杞多糖,每天1次,连续8周,安慰剂对照组灌胃服用等量蒸馏水,空白对照组正常饲养,不做任何处理。应用老化度评分评估老化征象和 Morris水迷宫检测小鼠学习记忆能力。结果 ①3、5、7 月龄SAMP8组与同月龄SAMR1对照组比较:3月龄SAMP8各组均较3月龄SAMR1对照组老化度明显增加,学习记忆能力明显下降(P<0.05),5、7月龄与3月龄结果相同; ②3、5、7 月龄SAMP8给药组与同月龄SAMP8空白和安慰剂对照组比较:3月龄SAMP8各给药组均较3月龄SAMP8空白和安慰剂对照组老化度减少、学习记忆能力提高,但差异无统计学意义(P>0.05); 5月龄枸杞叶高、低剂量组和枸杞多糖低剂量组均较5月龄SAMP8空白和安慰剂对照组老化度减小,学习记忆能力提高,具有统计学意义(P<0.05); 7月龄各给药组结果与5月龄相同; ③3、5、7月龄SAMP8各给药组间比较:5月龄SAMP8枸杞叶低剂量较3、7月龄各给药组老化度减小,学习记忆能力提高(P<0.05); ④同月龄SAMP8高、低剂量组间比较:3月龄SAMP8枸杞叶、枸杞多糖低剂量组均较3月龄高剂量组老化度减小,学习记忆能力提高,但无统计学意义(P>0.05); 5月龄SAMP8枸杞叶、枸杞多糖低剂量组均较5月龄高剂量组老化度减小,学习记忆能力提高(P<0.05); 7月龄与5月龄结果相同。结论 枸杞叶全汁和枸杞多糖对各月龄快速老化模型小鼠SAMP8的老化征象和学习记忆能力均有改善作用,但改善的程度与服用剂量及起始干预年龄有关。
Abstract:
Objective To investigate the effects of lycium barbarum leaves and lycium barbarum polysaccharides on appearance of aging sign and learning and memory in senescence- accelerated mice(SAM). Methods Lycium barbarum leaves(1, 10 mg·kg-1)or lycium barbarum polysaccharides(0.1, 1 mg·kg-1)were orally administered to 3, 5 and 7-month-old SAMP8 mice once a day for 8 weeks. Age-related changes were monitored by “the scores of aging”. Learning and memory performance were evaluated by the Morris water maze tests. Results The SAMP8 appeared aging sign and symptoms at 3-month old. Compared to SAMR1 control group,SAMP8 mouse showed significant impairment on learning and memory in hidden platform trialatage of the 3th,5th month and 7th month(P<0.05). This age-related learning and memory impairment was prevented by pretreatment with either Lycium barbarum leaves(1, 10 mg·kg-1)or lycium barbarum polysaccharides(0.1 mg·kg-1),P<0.05. Moreover, SAMP8 at 5th month with Lycium barbarum leaves(1 mg·kg-1)showed better effect than other group did. Conclusion These results suggested that lycium barbarum leaves and lycium barbarum polysaccharides could prevent the appearance of aging sign and symptoms,and age-related learning and memory impairment. These compounds may have potential therapeutic applications to various neurodegenerative disorders.
