摘要:
目的 应用生物信息学分析软件预测细粒棘球蚴中国大陆株诊断抗原P-29氨基酸序列,了解该蛋白的特性,预测其抗原表位,为P-29进一步应用研究提供理论依据。方法 应用DNAStar、BIOSUN、EXPASY/SignalP、protparam、psite/softberry生物分析软件对P-29的二级结构、抗原表位、信号肽、极性进行预测分析,用EXPASY/SWISSMODEL对蛋白的三维结构进行模拟。结果 P-29基因开放阅读框编码一个由238个氨基酸残基组成的多肽, 分子式为C1180H1906N324O386S9,其蛋白分子量为27095.58 Da,等电点为5.649,有37个碱性氨基酸,41个酸性氨基酸, 68个极性氨基酸和76个疏水性氨基酸,半衰期是30 h,结构稳定,呈亲水性且为带负电荷的酸性蛋白,α-螺旋水平较高,有15个抗原决定簇,无信号肽,有2个N-糖基化位点,6个蛋白激酶C磷酸化位点,11个酪蛋白激酶II磷酸化位点及7个微体羧基端目标信号位点。结论 根据生物信息学预测的结果表明,P-29可能是免疫诊断、药物作用及疫苗潜在的靶分子。
Abstract:
Objective To analyze and predict antigen epitopes of the recombinant protein EgP-29 and to provide the theoretical basis for further application research of EgP-29. Methods The two level structure, epitopes, signal peptide, polarity of EgP-29 was predicted by DNAStar, BIOSUN, EXPASY / SignalP, protparam, psite / softberry biological analysis softwares, using EXPASY / SWISSMODEL on protein three-dimensional structure simulation. Results P-29 gene open reading frame encoded peptides consisted of 238 amino acid residues. Its molecular formula was C1180H1906N324O386S9, its protein with a molecular weight of 27095.58 Da and an isoelectric point of 5.649. The peptide had 37 basic amino acids, 41 acidic amino acids, 68 polar amino acids and 76 hydrophobic amino acid, alpha helix higher level. It included 15 epitopes, no signal peptide and the half-life was 30 hours. Its structure was stable and it is a hydrophilic and negatively charged acidic protein. Conclusion P-29 may be potential target molecules for immune diagnosis, drug and vaccine.
