摘要:
目的 利用腺病毒载体介导外源性rHSG转染至蛛网膜下腔出血(SAH)大鼠的基底动脉(BA),观察SAH后各时相BA形态学变化规律。方法 SD大鼠60只随机分为四组:SAH组(A组,18只)、SAH+Adv-GFP组(B组,18只)、SAH+Adv-rHSG-GFP组(C组,18只)和正常对照组(D组,6只)。建立大鼠蛛网膜下腔2次出血模型,将重组腺病毒Adv-rHSG-GFP及空载腺病毒Adv-GFP分别转染至BA。设立4、7、10d 3个观测点, HE染色BA后测量其内径周长及血管壁厚度,观察血管结构改变。结果 A、B组各时相BA均有不同程度的痉挛、缩窄的表现; 主要为血管内径明显变小,管壁增厚,弹力纤维皱缩,向血管腔内隆起。C组各时间点血管腔及管壁变化不明显,无痉挛发生。不同时间点纵向比较采用单因素方差分析, C组与A、B 组相应时间点比较差异有统计学意义(P<0.05); A、B组各时间点横向比较差异无统计学意义(P>0.05)。结论 rHSG的过表达可以缓解SAH后血管壁痉挛的发生。
Abstract:
Objective Using adenovirus-mediated exogenous rHSG green fluorescent protein vectors(Adv-rHSG-GFP)infected rat basilar artery smooth muscle cells(VSMCs), to explore dynamic histological changes of vasospastic basilar arteries. Methods 60 Sprague-Dawley rats were randomly divided into SAH group(A, n=18), SAH+Adv-GFP group(B, n=18), SAH+Adv-rHSG-GFP group(C, n=18)and normal control group(D, n=6). SAH were developed with two times injections of arterial blood into cisterna magna. Adv-rHSG-GFP and Adv-GFP were transfected into the BA of rats. The BA were harvested at day 4, 7, 10 after experiments. BA were stained by hematoxylin-eosin to measure the inner diameter of the circumference of the vascular wall thickness for observing the vascular structural changes. Results BA at all times in A and B group showed varying degrees of vasospasm. It mainly showed that vessel diameter got smaller, wall thickening and elastic fiber shrinkage to endovascular uplift. There was no vasospasm in C group at all time points. Compared with group A, B, the difference in vessel diameter and wall thickness was significant(P<0.05)at the corresponding point in time. Conclusion The overexpression of rHSG can relieve spasm of the vessel wall after SAH occurrence.
