摘要:
目的 探讨黄芪注射液对大鼠脑出血灶周围凋亡神经元线粒体Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶和细胞色素C的影响。方法 将32只雄性SD大鼠随机分为黄芪治疗组、出血模型组和正常对照组,其中黄芪治疗组又分为出血后6h治疗组和24h治疗组。选用胶原酶Ⅶ-肝素液注射到大鼠尾状核建立脑出血动物模型,48h后测定各组大鼠脑出血灶周围神经元线粒体Na+-K+-ATP酶和Ca2+-Mg2+-ATP酶的活性,以及神经元内细胞色素C的改变。结果 与正常组比较,出血模型组细胞色素C的表达明显增加(P<0.01),ATP酶的活性均明显降低(P<0.01)。与出血模型组比较,6h治疗组及24h治疗组的细胞色素C表达均明显减少(P<0.05),ATP酶的活性均明显增加(P<0.05); 6h治疗组与24h治疗组比较,细胞色素C的表达差异无统计学意义(P>0.05),ATP酶的活性6h组高于24h组(P<0.05)。结论 黄芪注射液进行干预可减少脑出血灶早期神经元细胞内细胞色素C的释放及提高Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶的活性,从而减轻脑出血灶区神经细胞的凋亡,并且出血6h用药较出血24h后用药效果更好。
Abstract:
Objective To investigate the effect of radix astragali injection on expression of Cytochrome C(CytC)and activities of mitochondrial Na+-K+-ATPase and Ca2+-Mg2+-ATPase in peri-intracerebral hemorrhage(ICH)foci of rats. Methods 32 male rats were randomly divided into the radix astragali remedial group and the contrast groups including ICH contrast group and normal contrast group without ICH. The radix astragali remedial group were divided into 2 groups which were 6h and 24 h remedial group. All rats involved in the experiment were killed on the 2nd d after ICH. The cerebral hemorrhage rat model was established by injecttion collagenase Ⅶ-heparin solution into the caudate nucleus. The expressions of CytC and activity of ATPase(Na+-K+-ATPase and Ca2+-Mg2+-ATPase)were detected after two days. Results Compared with the normal contrast group, the expression of CytC in the control group with ICH increased significantly(P<0.01)and the activity of ATPase decreased significantly(P<0.01). Compared with the ICH contrast group, the expression of CytC in the radix astragali remedial group decreased significantly(P<0.05)and the activity of ATPase increased significantly(P<0.05). Compared two radix astragali remedial groups(2d-6h and 2d-24h), no significant difference were on the expression of CytC(P>0.05). The activity of ATPase at group 2d-6h was significantly higher than that at group 2d-24h(P<0.05). Conclusion radix astragali injection can relieve the release of neuron CytC and increase the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in peri-intracerebral hemorrhage foci, thereby to lighten the apoptosis of ICH. It has better effect to inject radix astragali earlier.
