摘要:
目的 探讨死亡受体5(death receptor 5,DR5)在颈椎间盘退变中的作用。方法 采用原位DNA片段末端标记( TUNEL)法和免疫组织化学方法,对比观察30例颈椎病患者(实验组)及30例创伤患者(对照组)颈椎间盘组织细胞凋亡状态及死亡受体DR5的表达。结果 TUNEL染色显示实验组的髓核内平均凋亡细胞率为(10.93±1.87)%,高于对照组的(6.43±1.56)%(P<0.01)。免疫组织化学染色显示实验组的髓核内DR5阳性细胞率为(50.73±15.92)%,高于对照组的(41.23±16.35)%(P<0.05)。Pearson相关分析表明颈椎间盘髓核内DR5阳性细胞率与TUNEL阳性细胞率之间呈正相关(r=0.755,P<0.01)。结论 高水平的DR5表达参与了退变颈椎间盘髓核细胞的凋亡,由DR5/TRAIL诱导的凋亡途径可能在颈椎病颈椎间盘退行性变发生和发展中发挥作用。
Abstract:
Objective To explore the expression of death receptor 5(DR5)in degenerative cervical disc tissue.Methods The apoptotic status andthe expression of DR5 were detected in 30 cases of degenerative cervical intervertebal disc tissues and 30 of traumatic normal cervical intervertebal disc tissues with TdT-mediated dUTP-biotin nick end labeling( TUNEL)and immu- nohistochemistry methods.Results The positivity rate of TUNEL staining was significantly higher in patient group(10.93%±1.87%)than that in control group(6.43%±1.56%)(P<0.01).The positive expression rate of DR5 of nucleus pulposus in patient group(50.73%±15.92%)was significantly higher than that in control group(41.23%±16.35%)(P<0.05).When the total 60 specimens in the two groups were analyzed,the DR5 positiveincidence of nucleus pulposus showed significant positive correlation with theTUNEL positive incidence of nucleus pulposus(r=0.755,P<0.01).Conclusion High levels of DR5 expression in degenerative cervical intervertebal disc tissues is involved in apoptosis.The apoptosis pathway induced by DR5/ tumor necrotic factor-related apoptosis-inducing ligand( TRAIL)maybe play an important role inthe occurrence and progression of cervical intervertebal disc degeneration.
