摘要:
目的 探讨缝隙连接蛋白43(Connexin 43,Cx43)在缺血预处理心肌保护中的作用和地位。方法 将48只SD大鼠分为假手术组(S组)、缺血再灌注组(IR组)、缺血预处理组(IP组)和5-羟基癸酸+缺血预处理组(5-HD + IP组),测定各组的心律失常评分和心肌梗死面积,采用逆转录-聚合酶链反应技术(RT-PCR)检测各组心肌Cx43的表达水平。结果 IR组和5-HD+IP组的心律失常发生多于IP组,评分明显高于IP组(P<0.01); IR组和5-HD+IP组的IS/AAR比值明显大于IP组(P<0.01); 与IR组和5-HD+IP组比较,IP组Cx43 mRNA的表达明显增强(P<0.01)。IP可以减少心律失常发生,减小心肌梗死面积,增强Cx43的表达,5-羟基癸酸可以阻断IP的心肌保护作用。结论 IP通过开放线粒体ATP敏感性钾通道保持Cx43磷酸化,进而实现心肌保护。
Abstract:
Objective To investigate the effect and status of connexin 43 on myocardial protection in ischemic preconditioning.Methods 48 Sprague-Dawley rats were randomly divided into four groups: S,IR,IP,5-HD+IP.The myocardial infarct size and arrhythmia score were measured.The expression of connexin 43 was detected by RT-PCR.Results Compared with group IP,arrhythmia score and IS/AAR was higher and the expression of connexin 43 was notablely lower in group IR and group 5-HD+IP(P<0.01).Ischemic preconditioning might reduced arrhythmogenesis and myocardial infarct size,meanwhile enhanced the expression of connexin 4 3.5-hydroxydecanoate acid might interdicted the myocardial protection of IP( P<0.01).Conclusion Ischemic preconditioning might play a n important role in myocardial protection by activating mitochondrial ATP sensit ive potassium channel and preserving connexin 43 posphorylation.
