摘要:
目的 研究新型格列酮类化合物ANY2对原代大鼠前体脂肪细胞糖脂代谢的影响。方法 ①用Ⅱ型胶原酶消化法得到原代大鼠前体脂肪细胞接种于24孔细胞培养板,以诱导培养基诱导分化,油红O染色法观察化合物ANY2对前体脂肪细胞分化的影响,并用异丙醇萃取油红O测定吸光度,比较各组分化程度。②以高浓度地塞米松作用于分化的脂肪细胞,制备胰岛素抵抗模型,观察化合物ANY2对胰岛素抵抗脂肪细胞葡萄糖消耗量(GC)和游离脂肪酸(FFAs)溢出量的影响。结果 浓度为1.1mg·L-1的化合物ANY2能显著促进前体脂肪细胞分化,分化率高达152%,提高其储脂能力,增加小脂肪细胞数量; 提高胰岛素抵抗脂肪细胞糖耗量达363mg·mg pro-1,减少其游离脂肪酸溢出,溢出量仅69 mmol·mg pro-1。结论化合物ANY2可促进原代前体脂肪细胞分化充脂,并改善高浓度地塞米松所导致的胰岛素抵抗作用。
Abstract:
Objective To investigate the effects of a novel thiazolidinedione ANY2 in primary precursor adipose cells.Methods 1.Using enzyme-digesting method,primary precursor adipose cells from the rat adipose tissues were cultured in 24wells Culture Plate.Using culture medium to induce the transformation of the cells and stained with oil red O to contrast the differentiation.2.Differentiaedcells were exposed to high concentration of dexamethasone to build model of Insulin resistance cells.The effects of ANY2,including glucose consumption(GC)andFFAs releastion were tested in this model.Results ANY2 could significantly promote the differentiation on primary precursor adipose cells,increase consumption of glucose[363mg·(mg·pro)-1] and reduce FFAs releastion[69 mmol·(m g·pro)-1)]underthe state of insulin resistance.Conclusion Compound ANY2 has remarkable effects on improving insulin resistence in adipose cells.
