摘要:
目的 观察甘草酸对糖尿病肾病大鼠肾小球α平滑肌肌动蛋白(α-SMA)表达的影响,探讨甘草酸防治糖尿病肾病的作用及其可能机制。方法 正常雄性SD大鼠随机分为正常对照组(简称对照组)、糖尿病模型组(简称模型组)、糖尿病甘草酸干预组(简称干预组)。采用腹腔注射链脲佐菌素制备糖尿病大鼠模型。干预组给予甘草酸灌胃(100mg·kg-1),每天1次,分别注射14、30和60d。对比观察14、30、60d肾脏病变,电镜观察肾小球变化,免疫组织化学方法观察肾小球α-SMA的表达。结果 光镜和电镜显示,对照组肾小球结构正常,与糖尿病组相比,甘草酸组肾小球系膜区的宽度小于糖尿病组,可见系膜、基底膜等病变减轻。免疫组化可见,60d时,甘草酸组大鼠肾小球α-SMA相对含量(2.65士0.20)较糖尿病组(3.30±0.45)降低(P<0.05)。结论 甘草酸对糖尿病大鼠肾小球具有一定的保护作用,甘草酸组肾小球α-SMA表达减少可能与甘草酸的作用有关。
Abstract:
Objective To observe the effect of glycyrrhizin in preventing and curing diabetic nephropathy and to explore the influence of glycyrrhizin on expression of α-SMA of glomerulus. Meanwhile, to approach the effect and possible mechanisms of glycyrrhizin on preventing diabetic nephropathy. Methods The normal SD rats were randomly divided into normal control group, model group, diabetic rats treated with glycyrrhizin group. Models were established by intraperitoneal injection of STZ. Diabetic rats treated with glycyrrhizin group were filled with glycyrrhizin once a day(100 /kg). The kidney changes were observed on the 14th day, 30th day, and 60th day by contrasting and the expression of glomerulus α-SMA were observed by immunohistochemistry,and the kidney structure changes were demonstrated by electronic microscope. Results Normal morphology were found in the control group in the light microscope and electronic microscope. Compared to the diabetic groups, the width of the glomerular mesangium of the glycyrrhizic treated group was significantly smaller and the pathological changes of mesentery and basilar membrane were alleviated. On the 60th day, compared with the diabetic groups(3.30±0.45), the relative amount of α-SMA in the glomerulus(2.65±0.20)was decreased(P<0.05). Conclusion Glycyrrhizin is able to protect the glomerulus from diabetic nephropathies. Glycyrrhizin can inhibit the expression ofα-SMA in diabetes glomerular mesangium.
