摘要:
目的 探讨癫痫对大鼠卵巢和生长卵泡的影响。 方法 双极电极植入大鼠左侧杏仁核,给予连续电刺激,建立电点燃的癫痫模型; 假手术组,电极植入后不给予电刺激; 对照组为正常雌性大鼠。癫痫模型大鼠在点燃20d后于动情间期取双侧卵巢,同期其它两组大鼠也分别于动情间期摘取双侧卵巢,右侧卵巢HE染色后光镜下阅片,通过Image-Pro Plus 6.0软件对窦前卵泡、窦卵泡、闭锁卵泡及卵泡总数进行分析,左侧卵巢选择窦状卵泡进行透射电镜观察。 结果 癫痫模型组大鼠卵巢增大,卵巢中窦前卵泡、总卵泡数及闭锁卵泡显著增多(P<0.05),窦状卵泡颗粒层变薄,卵泡膜明显增厚,电镜下显示癫痫组大鼠窦状卵泡中的颗粒细胞凋亡增多,增生的卵泡膜细胞更加富含脂滴。 结论 癫痫可导致大鼠卵巢发生多囊性改变,生长卵泡凋亡增多,卵泡膜细胞增生分泌增强。
Abstract:
Objective To explore effect of amygdale-kindled seizures on ovarian and growing follicles in female rats. Methods Kindling electrodes were implanted in the left basolateral amygdala of adult female rats. The rats were kindled by brief, daily, suprathreshold stimulations with a bipolar electrode. The sham-kindled rats only implanted bipolar electrode in the left basolateral amygdala. Controls were normal female rats. Electrographic and behavioral seizures were monitored. Ovary tissues were taken after 20 days of kindled in the diestrus. At the same time, sham-kindled subjects and their yoked controls were sacrificed and their ovaries were taken in their diestrus. Ovarian morphometric and growing filliches analyses were performed. Results The result of HE staining showed that: compared with control group and shamed-kindled group, kindled female rats had enlarged ovaries with numerous growing follicles, espically many preantral follicles, and significantly more follicles and atretic follicles(P<0.05); vesicular follicle showed more thinner granulose cell layer and thicker theca cell layer in kindled female rats. Ultrastructure of vesicular follicles of kindled rats showed increase of apoptotic granulose cells. More secretory granules could be found in theca cells and their ovarian exhibited polycystic changes. Conclusion Seizures induced by amygdala can result in polycystic ovary,more apoptosis in growing filliches and increase of the secretion in the theca cell hyperplasia.
