摘要:
目的 观察CyclinE在食管鳞状细胞癌和癌周正常食管组织的表达,从量化角度阐明其在食管鳞状细胞癌组织中表达的病理学意义。方法 常规石蜡包埋、HE切片确诊,免疫组化MaxVision法检测CyclinE在食管鳞状细胞癌和癌周正常食管组织的定位和表达,Image-pro plus图像分析软件对其表达强度进行定量分析,并用表达的阳性单位(positive unit,PU)反映其表达强度。结果 CyclinE蛋白在食管鳞状细胞癌和癌周正常食管组织中的表达主要定位在细胞核。食管鳞状细胞癌表达的PU值高于癌周正常食管组织(P<0.01),CyclinE蛋白表达的PU值与食管癌分化程度有关(P<0.01),低分化鳞状细胞癌表达的PU值高于高分化及中分化鳞状细胞癌。CyclinE蛋白表达的PU值与食管癌患者的性别、年龄、转移和TNM分期未见明显相关性(P>0.05)。结论 CyclinE蛋白表达程度与食管鳞状细胞癌及分化程度具有相关性,可能成为判断其指标之一。
Abstract:
Objective To observe the expression of CyclinE in esophagus squamous carcinoma and the normal esophagus tissue and to clarify its significance in esophagus carcinogenesis. Methods CyclinE protein expression was detected by immunohistochemistry(MaxVision method)using the monoclonal antibody and its intensity was assessed quantitative by image-pro plus image analysis system. Results The monoclonal antibody showed specific cell nucleus. CyclinE positive unit( PU)were significantly larger in the esophagus squamous carcinoma than that in normal esophagus tissue(P<0.001).CyclinE PU of the nuclei were significantly larger in the poorly-differentiated esophagus squamous carcinoma than those in the highly-differentiated esophagus squamous carcinoma and the moderately-differentiated esophagus squamous carcinoma(P<0.001). CyclinE PU was not involved in the patient's sex, age, tumor'metastasis and staging(P>0.05). Conclusion CyclinE protein expression was involved in the esophagus carcinogenesis and its differentiation and might be helpful for distinguishing carcinoma from normal tissues and ascertaining the histological grading.
